OROBOROS FAQ



1. Q: Got confused with o2k-core and o2k-module? If I understood it right, o2k-core if purchased can be converted in future to measure other functions for example fluorescence, Ca+2, pH.


A: The O2k-Core is the actual instrument with the accessories, syringes and the ISS suction system. With the O2k-Core you can measure respiration, with the O2k-Modules you are able to measure different parameters as well. The Modules are ready to use with the O2k-Core without any special upgrading needed.

2. Q: How about installation of the machine in the lab here in US? Is anybody going to do it for us? Or, buyer has to do it.


A: Set-up and installation is performed by the customer with the help of our detailed and userfriendly O2k-Manual. Our customers from all over the world have been finding this very easy and our Manual is also freely available for download online. Please see: http://www.oroboros.at/index.php?o2k-manual. The manual, as well as the troubleshooting section on our wiki-based website (http://www.bioblast.at/index.php/Troubleshooting) and our technical specialists help to locate and solve any technical problems that might appear. Any parts required for repair onsite are shipped to the customer. For any grave technical faults which cannot be solved on site, the O2k is shipped to our workshop, where it is repaired.

3. Q: What is the advantage of titration-injection micropump? Is manual pipetting to add sample to the chamber difficult? How is it different from ISS-integrated suction system and microsyringe set in O2K-core?


A: TIP2k: Basically, the TIP2k makes life easier and work more comfortable, like automated determinations of oxygen background. If you are interested in e.g oxygen kinetics at low oxygen or very precise small step uncoupler titration, then the more advanced features of the TIP2k are necessary. If you want to use the O2k-Modules mentioned above were strongly recommend purchasing also the TIP2k. Most MultiSesnor measurements are also possible without it, but it gets dedious. ISS: The ISS is for siphoning off excess medium from the O2k-Stopper and for emptying the O2k-Chambers. The ISS is included in the purchase of an O2k-Core and this suction pump is essential for the everyday usage of the instrument. The Microsyringe Set is another O2k-Core-accessory used for manual titrations.

4. Q: Are fluorescence sensors used for only green and blue emission? Is sensor not suitable for red emission? How about the excitation? Does the machine use a wide excitation spectrum?


A: You are correct, there was a confusion of excitation and emmision on our webpage. I have corrected this on http://wiki.oroboros.at/index.php/O2k-Fluorescence_LED2-Module. Thank you for bringing this to our attention!. Please note that the applications currently envisaged are listed on the top of the linked page (under description). We do not use a wide exciation but separate LED sources for the two mentioned excitation colors.

5. Q:If only Ca+2 is detected with the suitable module? Or, other ions such Na+, phosphate, K+ can also be detected.

6. Q: Is special organ tissue or cell preparation needed to measure ions? Or, whole tissue and adherent/suspended cells are used.


A: Adherent cells (without bringing them in suspension are conceptional not well suited for our system, see http://wiki.oroboros.at/index.php/Adherent_cells. TPP+ has been measured now with three types of samples, isolated mitochondria, permabilized cultures cells and permeabilized tissues. I do not see any reason that would exclude a type of sample here.

7. Q: Where do you get NO sensor and the cost of it?


A: NO-Sensors: NO and H2O2 sensors use an amperometric measurement principle (Amp-Module), comparable to the polarographic oxygen sensor (POS), which yields an amperometric signal at a given polarization voltage. We supply custom-made stoppers that can accommodate such sensors. All amperometric sensors, like (H2S, H2O2) that have the same connection, can be used with the O2k. However, we have to mention that, while NO is measured by quite a few of our customers, the usability of any H2O2 electrode currently on the market is sometimes questioned. However, this depends on your sensitivity needs. We offer the O2k-MultiSensor Stoppers, for further information please check the following link: http://www.bioblast.at/index.php/O2k-NO_Amp-Module. For prices of NO electrodes (shaft with 2 mm diameter) please consult the manufacturer, e.g. WPI. WPI's 2 mm electrode is called "ISO-NOP" You will need the electrode housing , "replacement" sleeves #5436 (there are non with just the housing) and filling solution #7325. That is, unless they have changed the composition of their packages, so please inquire with them. For compatibility with our system the important point is the diameter of the sensor: 2mm.

8. Q: How are we going to avail parts and labor warranty in the US?


A: With our general terms and conditions we have the following warranty: 24 months* for parts and labour from day of delivery, except glassware and consumables. (*This warranty period applies in conjunction with participation in an Oxygraph-2k Course within 12 months of delivery. Otherwise the warranty period is reduced to 12 months.). To this, we can ship spare parts to you if you require any and if there is anything serious with the instrument it can be shipped to our Workshop.

9. Q: Please explain Delivery included but duty unpaid? I do not get it.


A: DDU (Delivery included - Duty Unpaid) means: We pay for the shipment (delivery) of your order and you pay for the local VAT and the customs duty for the import of the instrument.

10. Q: Different module mentioned includes o2k-chamber and oxygen sensor as spare. Do these go bad with usage? Do you recommend to have them as a backup?


A: O2k-chamber and Oxygen sensor: It is not required to have these items as backup however as they are quite essential to the usage of the instrument we recommend having them as spares. As the O2k-Chamber is out of glass, it may be good to have a backup, but not necessary. This is completely up to you. The oxygen sensor (POS) has a two year warranty and we recommend regular sensor maintenance to keep its lifetime to a maximum. POS maintenance: The sensors do not need to be shipped. 11. Q: With regard to workshop: If O2K-fluorescence is different from O2K-basic. What will be covered in the workshop in Sydney, Australia? Is registration fee included with the purchase for the workshop in Sydney?


A: The registration fee which is included with the purchase of the O2k-Core is valid for any O2k-Basic-Workshop. The Workshop in Sydney is an O2k-Basic-Workshop so you are free to go there if you prefer this destination. The registration for the Fluorescence-Workshop comes with the purchase of the Fluoresence-Module.
