Description
Octanoylcarnitine is a medium-chain fatty acid (octanoic acid: eight-carbon saturated fatty acid) covalently linked to carnitine, frequently applied as a substrate for fatty acid oxidation (FAO) in mitochondrial preparations.
Abbreviation: Oct
Reference: pdf
MitoPedia topics: Substrate and metabolite
Application in HRFR
Oct: Octanoylcarnitine (DL-Octanoyl-carnitine-HCL, C15H30NO4Cl); TOCRIS Bioscience, Cat. No. 0605, 50 mg, store at R.T, desiccate); FW = 323.85.
Alternative: (±)-Octanoylcarnitine chloride, BioTrend, Cat. No. BS0136, 50 mg, store desiccate at RT; FW = 323.86.
Caution: Chemicals stored in the fridge or freezer should be allowed to reach room temperature before opening.
Preparation of 100 mM stock solution (dissolved in H2O):
- Weigh 50 mg of octanoylcarnitine and dissolve in 1543 µl H2O.
- Divide into 0.2 ml portions.
- Store at -20 °C.
O2k manual titrations into 2 ml O2k-chamber
- Titration volume: 4 µl using a 10 µl syringe.
- Final concentration: 0.2 mM in MiR05 or MiR06. Optimum Oct concentrations have to be evaluated, particularly in media with BSA concentrations different from MiR06.
