- Fig. 1.5. Partitioning of a sample of suspended cells of suspension volume Vs [mL]. (1) vs1 for cell count Nce [Mx = 106 x]; (2) cell-count concentration Cce [MxโmL-1]; (3) vs2 for cell wet mass mce [mg] or other markers of cell size and mt-markers; (4) mass per elementary cell MUce [mgโMx-1 = ngโx-1]; (5) vs3 or Vstock for the respirometric stock of suspended cells; and (6) vstocki subsamples for technical repeats of respirometric chambers. Typical numerical values are given for (a) human umbilical vein endothelial cells HUVEV or fibroblasts, and (b) cryopreserved HEK 293 cells. The targets are a sample-cell suspension with a cell-count concentration of 10 to 40 MxโmL-1; a final cell-count concentration of 1 MxโmL-1 in the respirometer chamber, which may be reduced to 0.5 MxโmL-1; a subsample titration volume of vstocki = 0.1 mL to 0.5 mL into the 0.5- or 2.0-mL experimental chamber; and a wet cell mass of about 1 mg for gravimetric measurement or other assays. For HUVEC with a radius of 6.8 ยตm of the spherical suspended cells, the elementary cell volume VUce is 0.0013 nLโx-1 (1300 fLโx-1; Gnaiger 2003), and at a cell density of 1.06 ยตgโnLยญ-1, the elementary cell mass MUce is 1.38 mgโMx-1 (equivalent to ngโx-1).
