It is challenging to isolate from cultured cells enough mitochondria for high-resolution respirometry. Then you will always wonder how the mitochondria were affected by the isolation and whether you obtained a representative fraction. Instead we developed techniques for high-resolution respirometry with the OROBOROS O2k of neuronal mitochondria in situ, with mitochondria in their normal environment inside neurons attached to a substrate. Here are our results:
